چکیده

The aminopeptidase specificity of 24 strains of bacteria was determined fluorometrically by use of a series of alpha-amino acid beta-naphthylamides as substrates. Provided that strict control over medium and growth time was adhered to, a reproducible profile of aminopeptidase activity was obtained which could be used for the identification of bacteria.

Simultaneous multiple substrate tag detection with ESI-ion trap MS for in vivo bacterial enzyme activity profiling.

A bacterial identification method in which multiple enzyme activities are measured simultaneously and in vivo with electrospray ionization-mass spectrometry (ESI-MS) is described. Whole-cell bacteria are immobilized onto a filter support and incubated with a mixture of substrates. Each substrate is chosen to measure a specific enzyme activity of a targeted bacterium and to produce a tag of uniq...

Breakdown of peptides from a casein hydrolysate by rumen bacteria. Simultaneous study of enzyme activities and physicochemical parameters.

The breakdown of a pancreatic hydrolysate of casein (tryptone) by an inoculum of ruminal mixed bacteria was studied in vitro. Peptides were degradated at 33% after 5 h. The dipeptidyl aminopeptidase type 1 (DAP-1), exoaminopeptidase and leucine aminopeptidase (LAP) like activities were measured using, respectively, Gly-Arg-MNA, Ala-pNa and Leu-pNA as substrates. While the total proteolytic acti...

New peptide deformylase inhibitors and cooperative interaction: a combination to improve antibacterial activity.

OBJECTIVES Bacterial drug resistance is a worrying public health problem and there is an urgent need for research and development to provide new antibacterial molecules. Peptide deformylase (PDF) is now a well-described intracellular target selected for the design of a new antibiotic group, PDF inhibitors (PDFIs). The initial bacterial susceptibility to an inhibitor of a cytoplasmic target is d...

Activity of Dipeptidyl Peptidase-IV/CD26 and Aminopeptidase N/CD13 in Secretome of Mesenchymal Stem Cells after Treatment with LPS and PMA

Background: Emerging evidence suggests that secretome of mesenchymal stem cells has many anti-inflammatory and regenerative properties, which makes it a suitable candidate for the treatment of autoimmune and degenerative diseases. Dipeptidyl Peptidase-IV (DPP-IV)/CD26 and Aminopeptidase N (APN)/CD13 are ubiquitous ecto-enzymes which can digest various substrates including some ...

A rapid method for the estimation of the microbiological quality of refrigerated raw milk based on the aminopeptidase activity of Gram-negative bacteria

A rapid method for the estimation of the microbiological quality of refrigerated raw milk was developed based on the aminopeptidase activity of Gram-negative psychrotrophic bacteria, the dominant microbial population in refrigerated foods. Two versions of the test for measuring the aminopeptidase activity are reported (direct and indirect). A good correlation (r=0.93–0.95) between bacterial cou...
